The most important mechanic that governs the Roathus Lagoon which will let you get around the place fast is the Queen Anne boost. Every time you walk in high grass, the local Anklegator dictator will pop out, dealing damage and knocking the Kid into the air(she's only trying to help). Since we can move in mid-air by rolling, this can be heavily abused to skip walls and move over big gaps. The big thing to remember is that the boost has a 'cooldown' - it can only happen once each few seconds. In addition, the first time it becomes available will be a couple seconds after reaching the area with the Brusher's Pike.

From the Brusher's Pike platform, there are 3 gates(black rock walls) that you have to get through to reach this level's boss.

First gate
The way you will skip this gate depends only on your game mode:
NG - Since you picked up the Pike, the boost will be available soon. Roll into the grass on your right and walk around until you get hit by Anne. As soon as you can, roll straight right. This might be slightly hard at first, but with enough distance you should make it over the gap to platform number two.
NG+ - Ignoring the pike means you have to wait longer for Anne to be able to boost you. Waiting is bad in a speedrun, so walk over to the left side of the gate. You can walk between it and the skull, since there is no invisible wall there(it can even be done without rolling). Watch out for the ground behind, it forms quite slowly in some places.
Second Gate
There are again two ways to get through this one, but you can do either, in any category.
Method A - as you enter the platform, on the left side, a Pincushion will spawn. Kill it ASAP - in NG pike, musket shot, pike(in NG ASL, you might be able to use a grenade instead), in NG+ one musket shot up close is enough to kill it. If you did it fast enough, the gate will start opening and you will be able to roll under it. IF you were too slow, a second Pincushion will have spawned on the right side, preventing the gate from opening. In this case you will have to kill this one as well.
Method B - take a boost from the grass in the top right, and roll around the gate in mid-air from the top side.
Third Gate
Here, you do not take the intended way through the level(top-right). Instead, you will either clip or boost around the wide gate at the bottom.
Boost - Only do this boost if you didn't take the one before the second gate, since then you would have to wait. Stand in the small patch of grass over the left side of the rock wall to get it. This boost is the trickiest one because the gate's invisible wall extends really far left. To get around, aim your mid-air recovery down and a bit to the left, then, once you are below the gate, roll straight right. If you fall, don't bother trying to get another boost, go for the clip instead.
Roll Clip - This gate has a lot of holes in it that can be clipped through, but the fastest one is on its far right side, past the wall. It might take more than one roll, and requires some precise positioning(if you find this hard, you might want to go for a slightly easier but much slower clip inside the rocks on the right side; it's hard to explain, but requires changing your angle after you get inside the wall).

Queen Anne

Up until this point, Anne could not be damaged. After getting through the last gate, in the area with the Shard(that you can't pick up yet, but don't worry about it), as fast as possible move close to the black wall on the right side. This will begin the boss fight. At first Anne will start moving towards the Shard, and won't do anything else. During this time pick up the Shard if you don't need to be at low health to activate Werewhisky, or spend this time falling off to take damage and activate it instead. After she reaches a point somewhere near where the Shard was, she will start chasing the Kid and you will be finally able to damage her.

Damaging Anne is quite simple: Rolling over her makes her pop out for about two seconds, during which she is vulnerable to damage(avoid standing on her, as that will boost you instead). The way you will kill her depends on your weapon set and category, but you will usually aim for a 1- or 2-cycle. Each method is the same, just uses different attacks: roll over her, then quickly, and precisely, execute the relevant attack combination. This is challenging, and takes practice, but if you miss some attacks you can do more cycles without losing a lot of time at all. The basic thing to keep in mind is that you actually don't have to kill her - just deal enough damage to make Rucks say "Now he's made her mad". Doing this, or killing her, will activate the skyway letting you leave the level. Don't forget to grab the Shard if you were using Werewhisky.
